ELECTROPHORESIS ( REGULAR)

Electrophoresis is a separations technique that based on the mobility of ions in an electric field. Positivecharged ions migrate towards a negative electrode and negatively – charged ions migrate towards a positive electrode. Nucleic acids are routinely separated according to their mass and structural properties by Electrophoresis on Agarose gels. Liner double stranded DNA has a constant negative charge per unit length of molecule, resulting from the phosphate backbone of the helix. Thus, they migrate towards the anode during electrophoresis at a rate that is inversely proportional to the log of the molecular mass. Electrophoresis separation of nucleic acids therefore is strictly according to size. The molecular mass of double stranded DNA fragments can be determined after electrophoresis by comparison with standards of known molecular

Submarine (Horizontal) Gel systems

Submarine gels, often called Horizontal agarose gels, are routinely used in the laboratory to analyze DNA. Agarose gels are submerged in electrophoresis buffer in a horizontal electrophoresis apparatus. The buffer and gel form a current bridge between the two electrode chambers. Ethidium bromide is mixed in the gel matrix to enable fluorescent visualization of the DNA fragments under UV light after electrophoresis.

In addition to its importance as an analytical tool, gel electrophoresis is widely used for isolating and then purifying specific fragments of DNA, usually I preparation for sub cloning. Hence this is very commonly required procedure.

Cautions

  • Always check the wires for damage before using the unit and connect the cables to the powe supply BEFORE turning the power supply ON.
  • Do not use concentrated acids ,bases or halogenated and aromatic hydrocarbons.
    Never exceed maximum allowed voltage, current or power.
  • To avoid damage, use extreme care when cleaning this apparatus. After each use, rinse well with deionized water.
  • Wipe dry with soft cloths or paper towel, or allow to air dry.
  • Never use abrasive cleaners, window sprays, or scouring pads to clean the components, as these can cause surface damage.
  • Do not autoclave or dry-heat sterile the apparatus or components.
  • Do not expose the apparatus or components to phenol, acetone, benzene, halogenated
    hydrocarbon solvents, or undiluted laboratory alcohols.
  • Avoid prolonged expose of the apparatus or components to UV light.
